The aim of my project is the ultrastructural analysis of insulin secretory granules (SGs) of different age. Insulin secretion in pancreatic islets occurs in two phases: a short and rapid first phase followed by a sustained second phase. There is evidence of an age dependent release of SGs, i.e. “older” SGs are less likely to undergo exocytosis relative to “younger” SGs. I will use the latest techniques in fluorescence and electron microscopy to investigate the ultrastructure of SGs of different age and their interaction with the cytoskeleton of the beta cell as well the influence of SG markers on SG aging.
